Veuve du roi François II à seulement 18 ans, Marie veut récupérer le trône d'Ecosse, pays tiraillé entre catholiques et protestants. Mais sa cousine, la reine Elisabeth, va tout faire pour contrecarrer ses plans. Les deux reines s'affrontent par lettres interposées et combats armés.
