1942. Des milliers de réfugiés, venus des quatre coins de l'Europe, affluent à Casablanca, dans le fragile espoir d'obtenir un visa pour les Etats-Unis. Le Café américain leur sert de lieu de rendez-vous avec leurs contacts. Le propriétaire, Rick Blaine, est un homme secret au passé obscur, un individualiste farouche, cynique et désabusé. Le meurtre de deux émissaires nazis porteurs de lettres de transit conduit à Casablanca un important dignitaire allemand, le major Strasser. Peu après, le résistant roumain Victor Laszlo et sa troublante épouse, Ilsa, débarquent à leur tour au Café américain. Rick reconnaît en Ilsa la femme avec laquelle il a eu une liaison à Paris deux ans auparavant...
