Club president Sandro Rosell announced at a news conference that Tito Vilanova would step down from his role as coach of the Spanish champions due to a recurrence of throat cancer.
Rosell said a new head coach would be announced next week, and also that their first preseason friendly on Saturday - against Lechia Gdansk in Poland - would be cancelled.
44-year old Vilanova had to take two months off last season to receive treatment in New York.
In his single season in charge he led Barcelona to the Spanish league title with a record-tying 100 points.
